چکیده

The ability to hydrolyze starch is a property that is scattered among streptococcal species in 3 of the 4 physiological divisions described by Sherman (1937), the exception being members of the Enterococcus. A single reference to a starch hydrolyzing enterococcus is Mann's (Mann et al., 1954) report of a "starch-fermenting Streptococcus faecalis." Outstanding among the nonhemolytic streptococci for ability to hydrolyze starch is Streptococcus bovis and to a much lesser extent, Streptococcus equinus and some strains of Streptococcus uberis. Streptococcus thermophilus is reported to attack this substrate "on a favorable medium" (Breed et al., 1957). Sherman (1937) noted also that a few strains of Streptococcus lactis gave a very slight reaction on starch. A renewed interest in the starch hydrolyzing streptococci has resulted from the fact that S. bovis, or organisms of similar characteristics, have been shown to be present in considerable numbers in the rumina of the cow and the sheep. This fact has increased thought and speculation concerning the role of the starch splitting streptococci in the digestion of starchy feeds and in the general economy of the ruminants. For a reaction that has served as an important reference point in the delimitation of species of streptococci (S. bovis and Streptococcus inulinaceus) there has been remarkably little effort to standardize either the test or the interpretation of results. It is possible that a reexamination of cultural conditions and methods of testing would uncover species more marked in starch hydrolyzing potential than those revealed by the usual empirical methods. In addition to being an inhabitant of the

Purification and properties of starch hydrolyzing enzymes in mature roots of sugar beets.

Mature roots of sugar beets, which accumulate large amounts of sucrose but not starch, nevertheless contained acid and neutral amylases, judging from their pH optima, as well as pullulanase. Acid and neutral amylases were partially purified by procedures including fractionation with ammonium sulfate, ion exchange column chromatography, and gel filtration. Acid amylase was classified as an exoam...

Kinetic modeling of simultaneous saccharification and fermentation of corn starch for ethanol production.

Fuel ethanol production, using a simultaneous saccharification and fermentation process (SSF) of native starch from corn flour, has been performed using Saccharomyces cerevisiae and a granular starch hydrolyzing enzyme. The quantitative effects of mash concentration, enzyme dose and pH were investigated with the use of a Box-Wilson central composite design protocol. Proceeding from results obta...

CONSTRUCTION OF PARTIAL GENOMIC LIBRARY OF Bacillus subtilis AND SCREENING FOR RECOMBINANTS PRODUCING αAMYLASE, PROTEASE AND LIPASE

Genomic libraries contain a clone of every gene and extragenic sequences found in the cells of any organism and screening of the library is done to isolate clones that contain our gene of interest. Genomic libraries can be retained for many years and propagated and therefore it is a popular method of choice for maintaining every gene sequence of the organism whose gene library is being created....

The Differentiation of Three Distinct Desoxyribonucleases of Group a Streptococci

Group A streptococci were found to produce three different desoxyribonucleases, designated here as A, B, and C. These enzymes migrated separately on analysis with starch zone electrophoresis, were immunologically distinct, and showed certain differences in inhibition by citrate and in optimal pH of activity. The relative amount of each enzyme produced was a function of strain variation.

Detection and identification of group B streptococci by use of pigment production.

Pigment production by group B streptococci (GBS) is a useful test for identification of the organisms. The test is positive in 99.5% of beta-haemolytic strains. No false-positives are noted. Non-haemolytic strains do not produce pigment. Islam's media less agar can be used as a one-step broth detector of GBS in mixed cultures. This may have application for the detection of GBS in women in labou...
